ASC-RT | A Combined Raman and Microwave System: Uncovering Intermediate Components during Synthesis

A combined microwave and Raman system offers the possibility of controlled heating together with in situ reaction monitoring. To follow a reaction’s progress with Raman spectroscopy helps to reveal intermediate components and to gain more information about the actual reaction mechanisms.

To demonstrate the potential of Anton Paar’s combined microwave synthesis reactor and Raman spectroscopy setup, this report shows in situ monitoring of the synthesis of 3-acetylcoumarin. Coumarins are important natural compounds found in many plants with multi-biological activities (e.g. antibacterial, antioxidant, anticoagulant). They are widely used additives e.g. in perfumes, cosmetics, insecticides, and pharmaceuticals.

Here, they serve as a model system for a typical organic synthesis reaction in which a bicyclic structure is built up via an intermediate. Combining a Cora 5001 Raman spectrometer with a Monowave 400 R gives the opportunity to follow the intermediate in the reaction which usually only furnishes the product for further analysis but not the intermediate. Thus, reaction mechanism and kinetics studies are enabled leading to a better insight into the reaction processes. Consequently, the use of Anton Paar’s Raman and microwave combination setup paves the way to a fast and target-oriented process optimization.
