SAXS: A powerful tool for structural characterization of Biopharmaceutical samples

Six out of ten humans suffer from some stage of Chronic Kidney Disease (CKD), where selective filtration ability of their kidneys' get continually lost. There is no drug today which can directly cure this problem: either arrest the degradation or regain the loss. At physiological scale, it occurs due to effacement of finger-like cells called podocytes, which in turn is regulated by direct interaction between domains of two proteins: Cytoplasmic domain of Neph1 (Neph1CD) and First domain of PDZ ZO-1. For structure aided drug discovery, there is no crystal structure of this complex.

This talk will provide the insight of using how SAXS can help one to define the shape and how the speaker and his team has deciphered the shape of Neph1CD and its complex with binding proteins like ZO-1 and Myo1c.

Topics covered in the webinar are:

- The journey of using SAXS data and converging on the structure of Neph1CD
- The steps formulated to use this structure to discover a unique molecule, isodesmosine which "seals" this complex
- Journey towards validating results in cell lines, mice and zebra fish
- Finally, how Speaker and his group and collaborators are working together using SAXS to further develop lead molecule and identify more molecules to cure/manage the increasing problem of CKD.
